Dilab AccuSampler U Automated Sampling
The DiLab AccuSampler is a fully automated system for Automated Blood Sampling (ABS) in freely moving mice. The system handles sample volumes between 5 and 50 l and is optimized so that only minimum amount of blood (5-10 l) is borrowed from the animal during the sampling process. This makes it possible to take multiple samples from a mouse without any adverse effect to the animal. It is also possible to use the system for blood sampling on any other rodents where small sample volumes are required.
Traditional ABS systems borrow a comparatively large volume of blood (100 l+) from the animal, during the sampling process. Because of the small volume of blood that is available in a mouse (approx 300 l, or 10% of the total blood volume) this process can harm the animal as well as induce undesired stress responses. By virtually eliminating waste blood, the system can also be used for applications requiring smaller blood samples with a high degree of time resolution - as often as every 5 minutes to follow changes in drug levels or endogenous compounds.
In order to achieve accurate sample volumes the AccuSampler is equipped with a balance that weighs each sample individually directly after the sampling process. Once the sample is weighed, it is automatically chilled. All steps in the sampling procedures are monitored and logged by the AccuSampler Software. This allows the user to know the precise mass, and generate better quality analytical and PK data. In addition built in pressure sensors allow for the verification of the sampling process.
The animal is connected to the AccuSampler through a surgical catheter leading to a stainless steel swivel mounted on a balanced lever arm. The swivel makes it possible for the animal to turn without twisting the catheter. The animals can move freely during the study with access to food and water. Samples can be taken without disturbing the animal.
Two interconnected syringe pumps handle the blood sampling with great accuracy and precision. A patented, unique catheter design with a T-connector close to the mouse, in combination with the interconnected pumps, makes it possible to sample low volumes without any unwanted blood loss.
The AccuSampler is equipped with a 96 position rack for 100 l sample tubes. The collected samples can be cooled to a predefined temperature down to 0 C.
The sample vial is weighed on a built in balance, before and after the sample is taken and the sample weight is automatically determined. The blood weight is entered into the log file that is created for each process in the study and stored in the system database. The pressure sensor monitors the pressure in order to verify that the catheter is patent. The pressure values are also logged.
